Shares of ProKidney (PROK) soared more than 580% today following the release of encouraging topline results from its Phase 2 REGEN-007 clinical trial evaluating rilparencel, a cellular therapy designed to preserve kidney function in patients with chronic kidney disease and diabetes.
The trial demonstrated that patients receiving two scheduled rilparencel injections saw a statistically significant and clinically meaningful 78% improvement in the rate of kidney function decline. Specifically, the annual eGFR (a measure of kidney filtration rate) decline slowed from -5.8 to -1.3 mL/min/1.73m(2), with a p-value of less than 0.001.
A second group of patients received one injection and a second only if kidney function worsened. While this approach resulted in a 50% improvement in kidney function decline, the outcome was not statistically significant, suggesting a possible dose-response effect that may inform future studies.
Importantly, rilparencel was well tolerated across both groups, with no serious adverse events related to the treatment. The safety profile remained consistent with prior studies and comparable to that of a routine kidney biopsy.
These Phase 2 results are especially significant as they align closely with the design of ProKidney's ongoing Phase 3 PROACT 1 trial, which is already underway and aimed at securing accelerated approval in the U.S. using eGFR slope as a potential surrogate endpoint. The company expects to discuss this approach with the FDA during an upcoming Type B meeting.
Rilparencel, which has received the FDA's Regenerative Medicine Advanced Therapy designation, is being developed for patients with advanced CKD and diabetes, a group that currently faces limited treatment options beyond managing disease progression or eventual dialysis.
